Description

Intended use of the test kit

This diagnostic test is intended to identify antibodies against sugar antigens of Brucella melitensis, in serum and milk samples. In contrast to test systems which make use of agglutinating bacterial antigen, this partial monoclonal based ELISA has a very high sensitivity and specificity (according to SAT, E.C. and Weybridge standards).

Principle of the test kit

Diluted milk or serum samples are added to the pre-coated wells.

• Qualitative
  The serum sample is added (diluted 1:21) to the wells of the coated plate.
  The milk sample is added 1:1 diluted to the wells of the coated plate.
• Quantitative
  The serum sample also can be titrated using a 3-step dilution, starting with a dilution 1:30 (> 1:90 > 1:270 > 1:810).
  The milk sample also can be titrated using a 3-step dilution, starting with a dilution 1:1 (> 1:6 > 1:18 > 1:54)

After incubation and appropriate washing a monoclonal anti-ovine/caprine IgG antibody conjugate is added and the plates are again incubated. After appropriate washing, substrate is added. After several minutes the color reaction is stopped and the plates are immediately read at 450 nm.
